The Circulo de Bellas Artes is holding the event, which is set to start on Thursday and last for 48 hours, to mark the anniversary of Cervantes' death.
It did not reveal the exact time of the 14-year-old princess' participation.
The annual event, which is usually held in the Circulo de Bellas Artes' headquarters in central Madrid, will be held online due to the lockdown that was imposed to halt the spread of the novel coronavirus.
Infanta Sofia of Spain, Leonor's younger sister, who turns 13 later this month, is also to read an extract of Don Quixote, the organizers said.
Aside from the royals, politicians and other public figures taking part in the reading, members of the public including health professionals, police officers and supermarket workers will also be participating.
Spain's royal family has been completely absent from public life since the beginning of the coronavirus pandemic, which has hit Spain particularly hard. The family has come under fire from Spanish media as a result.
